A STimulated Emission Depletion (STED) nanoscopy platform is being implemented for plant histological analysis, which overcomes the light diffraction limit, reaching lateral and axial resolutions of 30-50 nm and 100-130 nm, respectively, allowing the visualization of live cells and tissues without compromising their integrity. Our Leica STELLARIS STED is equipped with white light (485-685 nm, in 1-nm path length) and diode (405 nm) excitation lasers, two STED (592 and 775 nm) lasers, a LIGHTING super-resolution module, and four hybrid spectral detectors.
